Mouse Anti-Human Bone Morphogenetic Proteins - 7
(Monoclonal Antibody)

Preparation:

Monoclonal antibodies were produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, E. coli-derived, recombinant human BMP-7.

Purification:

This IgG1 antibody was purified by Protein G affinity chromatography.

Formulation:

Lyophilized from a 0.2 um filtered solution in 0.5X phosphate-buffered saline (PBS) at pH 7.2.

Stability:

The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -800C.

Reconstitution:

When reconstituted in sterile water to a concentration of 0.5 mg/ml the antibody is stable for at least six weeks at 2-40C.

Applications

ELISA:

In a sandwich ELISA (assuming 100μl/well), a concentration of 1 μg/ml of this antibody will detect at least 0.5 ng/well of recombinant human BMP-7 when used with conjunction with goat anti-mouse horseradish peroxidase (Jackson Laboratories).

Western Blot:

To detect h-BMP-7 by Western Blot analysis this antibody can be used at a concentration of 2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant h-BMP-7 is 2.5 ng/lane.

Biological Activity:

To yield one-half maximal stimulation [ED50] of the biological activity of hBMP-7 (50 ng/ml), a concentration of 0.4 - 0.6 µg/ml of this antibody is required.
