Ella Principles:
The Ella automated ELISA system, developed by ProteinSimple, a Bio-Techne brand, is a sophisticated instrument designed to automate and simplify the enzyme-linked immunosorbent assay (ELISA) process.
The Ella Automated ELISA System: Advantages
The Ella platform, developed by ProteinSimple, automates the ELISA workflow, minimizing manual intervention and enhancing throughput. This system utilizes pre-loaded, single-use cartridges and microfluidic technology to perform assays within 90 minutes. Advantages include:
- Enhanced Throughput:
Traditional ELISA workflows are characterized by multiple manual steps, increasing the potential for variability and extending assay duration. Ella automates these steps, significantly reducing hands-on time and enabling higher throughput. Assays are completed within 90 minutes, a marked reduction compared to traditional methods. - Improved Reproducibility:
Manual ELISA procedures are susceptible to variability arising from differences in pipetting accuracy, incubation times, and washing efficiency. Ella's automated system ensures consistent and standardized assay conditions, resulting in improved reproducibility and reduced inter-assay variability. - Multiplexing Capabilities:
Traditional ELISA typically measures a single analyte per well. Ella facilitates the simultaneous measurement of up to eight analytes in a single run, conserving sample volume and reducing experimental time. - Reduced Sample and Reagent Consumption:
Traditional ELISA methods often require relatively large sample volumes. Ella's microfluidic technology enables assays to be performed with minimal sample and reagent volumes, particularly advantageous for limited sample availability. - Streamlined Data Acquisition:
Manual data analysis in traditional ELISA can introduce variability and prolong the analysis process. Ella integrates software for real-time data analysis, including curve fitting and concentration calculations, providing accurate and objective results. - Minimized Contamination Risk:
Traditional ELISA has a higher risk of well-to-well contamination due to the manual nature of the process. Ella uses single-use cartridges, therefore minimizing the risk of contamination.

Ella Automates and Accuracy
A high sample and a low sample are mixed in six different ratios to confirm the accuracy of this analyte with a constant plasma concentration.
Ella’s automation makes MCP-1 linearity studies much easier by reducing the heavy manual work involved in traditional ELISA (Fig. 1A). The same high plasma sample was tested across three different cartridges and resulted with excellent precision of 1.3% CV. Leading to much more accurate results (Fig. 1B)

Ella Reproducibility
This stability study for MIP-1beta evaluates the effect of freeze-thaw cycles on the cytokine's concentration. The recovery remains near 100% across multiple freeze-thaw cycles, indicating that the cytokine's concentration remains stable and unaffected. This ensures reliable results when retesting or using the sample in different testing protocols.
